What the business is
Biosciences startup developing tests and therapeutics based on CHIP, blood-cell mutations that indicate elevated risk for cancer and other age-related diseases, aiming to catch and treat them earlier
The bet
CHIP biology, understood for a decade but never operationalized by a company, is mature enough now to build therapeutics and diagnostics around
How it started
Mark Chao, a hematologist and former oncology VP at Gilead Sciences, had co-founded Forty Seven, an immuno-oncology company Gilead later acquired for close to $5 billion. In his clinical practice, he saw aggressive blood cancer patients referred to him only after multiple prior treatments, by which point their disease had already mutated too far for treatment to meaningfully help. CHIP biology — cell mutations that accumulate over a lifetime of roughly ten quadrillion (10^16) cell divisions and act as early markers for diseases like cancer — had been studied for about a decade, but Foresite Labs and Chao concluded no company had yet been positioned to operationalize it.
What happened
TenSixteen Bio launched in January 2022, co-founded with Foresite Labs, with a $40 million Series A from Foresite Capital and GV (formerly Google Ventures). Chao said the company would use the funding to grow from about 10 full-time employees to more than 30 over the following year while continuing to gather CHIP data, with the goal of having multiple treatments in clinical trials — or approved for oncology and heart disease patients — within five years.
What has to be true
A decade of published CHIP biology gave the company a scientific foundation without having to prove the underlying science from scratch
The founder's prior exit (Forty Seven to Gilead for ~$5B) on an adjacent precision-medicine thesis gave investors a track record to underwrite
Framing the company around treating disease earlier, before advanced mutation, matched a broader shift investors were already backing in diagnostics and precision medicine
What can be applied
A scientific thesis can sit understood-but-unbuilt for years; the opening isn't the discovery, it's finding the team and capital willing to operationalize it into an actual company.
