What the business is
Clinical-stage pharma developing D-PLEX100, a local-release doxycycline insert applied into the surgical wound to prevent infections for about 30 days.
The bet
D-PLEX is worth far more than the 2022 European license priced in — reclaim the rights and re-partner Europe as a de-risked, Phase 3-proven asset.
How it started
PolyPid licensed European commercialization rights to ADVANZ Pharma in 2022 for a €2.5 million upfront — a small slice of a package it valued at potentially more than $110 million — when its pivotal readout was still ahead of it.
What happened
SHIELD II, a 798-patient Phase 3 in abdominal colorectal surgery, met its primary endpoint and all key secondary endpoints, including a 60% relative risk reduction in surgical site infections (p=0.0013). The FDA accepted the New Drug Application with Priority Review, setting a November 28, 2026 PDUFA date; in July, Azurity licensed US and Canadian rights in a deal that has already delivered $30 million, with eligibility for roughly $290 million in milestones and tiered royalties. On October 1, 2026, the EMA validated PolyPid's MAA and the company regained exclusive European rights the same morning.
What has to be true
The 2022 deal carried a €2.5M upfront of a package valued at $110M+ — most value deferred to then-uncertain milestones.
Phase 3 success plus FDA Priority Review turned the same asset into a re-partnering opportunity on PolyPid's terms.
The Azurity deal validated the model: $30M already delivered, about $290M in milestones, tiered royalties.
EMA validation landed the same morning as the rights reclaim, keeping both major markets moving at once.
What can be applied
License early when you are weak, reclaim when you are proven: terms signed before pivotal data underprice the asset, and each regulatory win becomes negotiating leverage for the next deal.
Aftermath
As of October 9, 2026, D-PLEX is under regulatory review on both sides of the Atlantic, with the FDA decision due November 28, 2026 and an EMA approval — if granted — valid across all EU member states. PolyPid says it can now partner Europe on its own terms with a far more advanced asset than the one it licensed away in 2022.
